Effects of varied culture conditions on crude bacitracin produced by Bacillus subtilis isolated from the soil

Aim: To determine the effects subjecting Bacillus subtilis to different cultural conditions will have on crude Bacitracin produced in inhibiting growth of known susceptible organisms. Method: be a bacitracin producer was isolated from soil and identified. The isolate used produce under various culture with peptone water serving as basal fermentation medium. resultant checked for its antimicrobial activity against Staphylococcus aureus by using agar well diffusion method. Culture varied were addition glycerol, arabinose, fructose, mannose, sucrose, urea, MgSO4, CaCO3, KCl, sodium citrate, adjustment initial pH at 5, 6, 8 9. production done 37 oC 4 days an orbital shaker incubator rotating 150rpm. Result: test organism (Staphylococcus aureus) determined mm zone inhibition, best result obtained glycerol (14 mm) least inhibition observed MgSO4 (3 mm). Also, (20 mm), while reduced Conclusion: has effect aureus. highest activities can gotten raising broth that lowest effect.
